Ibrutinib

Ibrutinib is a kinase enzyme inhibitor with the anti-neoplasm effect that is prescribed in some blood disorders such as Mouttle cell lymphoma, CLL, SLL, Waldenstrom Macroglobulinemia, Marginal zene lymphoma, and chronic GVHD.

Ibrutinib prevents the activity of the protein that plays a role in the growth and spread of cancer cells in the body. Metabolism of this drug is through the liver and by CYP2D6, CYP3A4 enzymes.
The drug is mainly excreted through feces and part of it through urine.
Ibrutinib has a half-life of about 4 to 6 hours, and its peak blood concentration occurs 1 to 2 hours after consumption.
This drug has a very high protein binding (around 97%).

Mechanism of action

By irreversibly inhibiting proton tyrosine kinase (BTK), Ibrutinib prevents the function of B-cell antigen receptors and cytokines, and as a result, it can inhibit malignancies caused by abnormal proliferation of blood B cells.

Method of Use

The usual dosage of Ibrutinib is as follows depending on the individual’s conditions:

Patients with MCL and MZL: 560 mg once a day until disease progression or unacceptable toxicity.

– Patients with CLL, SLL, and WM: single drug or combination therapy (along with drugs such as bendamustine or rituximab): 420 mg once a day until disease progression or unacceptable toxicity

Patients with chronic graft-versus-host disease (cGVHD): 420 mg once a day until disease progression or return of underlying malignancy or unacceptable toxicity.
